2002年には『Leap of Faith』、2004年には『The D.A.』のレギュラーとなったが、どちらも数話で打ち切られた。映画では『恋は邪魔者』、『ハート・オブ・ウーマン』、『カーラの結婚宣言』、『Levitation』、『セレニティー』に出演した。
2006年から2007年にかけてはNBCの『Studio 60 on the Sunset Strip』のハリエット・ヘイズ（英語版）を演じ、ゴールデングローブ賞助演女優賞(シリーズ・ミニシリーズ・テレビ映画部門)にノミネートされた。
2008年にはウィル・アイズナーの同名コミックを映画化した『ザ・スピリット』でエレン役を務めた。
2008年8月25日、1998年の同名のテレビシリーズをリメイクした『Cupid』のパイロット版をABCが発注した。ポールソンはボビー・カナヴェイルの相手役を務めた。番組は2009年3月に始まったが、2009年5月19日に6話を以て打ち切られた。
2010年2月、『グレイズ・アナトミー 恋の解剖学』で若い頃のメレディス・グレイ（英語版）の母親役で出演した[5]。
2012年には2008年アメリカ合衆国大統領選挙を題材としたHBOのテレビ映画『ゲーム・チェンジ 大統領選を駆け抜けた女』でニコール・ウォレス（英語版）役を務めた。
2011年、FXのアンソロジーシリーズ『アメリカン・ホラー・ストーリー』の第1シーズン『呪いの館』に3話ゲスト出演した。翌2012年からの第2シーズン『精神科病棟（英語版）』ではメインキャラクターのラナ・ウィンターズ役で全話出演した。2013年からの第3シーズン『魔女団（英語版）』、2014年からの『怪奇劇場（英語版）』、2015年からの『ホテル（英語版）』、2016年からの『体験談（英語版）』、2017年からの『カルト（英語版）』、2018年からの『黙示録（英語版）』にも出演しており、本シリーズでの演技でエミー賞に5回ノミネートされている[6]。
2012年の映画『MUD -マッド-』、2013年の映画『それでも夜は明ける』、2015年には『キャロル』に出演した。
2016年、FXのミニシリーズ『アメリカン・クライム・ストーリー』の第1シーズン『アメリカン・クライム・ストーリー/O・J・シンプソン事件』にO・J・シンプソン事件で検察官だったマーシャ・クラークを演じ、エミー賞主演女優賞 (リミテッドシリーズ・テレビ映画部門)、ゴールデングローブ賞女優賞（リミテッドシリーズ・テレビ映画部門）等多数受賞。
