アメリカ合衆国のファーストレディ（アメリカがっしゅうこくのファーストレディ、英語: First Lady of the United States、略称: FLOTUS）は、通例アメリカ合衆国大統領（歴代全て男性）の配偶者である妻を指す語。ホワイトハウスのホステスであり、夫（配偶者）である大統領の補佐役でもあり、その多くは社会活動に携わっている。伝統的に、大統領が就任すると同時にその妻にこの地位が与えられている。

大統領が結婚していない（独身の）場合、または大統領夫人がファーストレディの役割を果たすことが不能とされた場合は、大統領がふさわしいと判断した女性にその代理を依頼する。
